Biological Background: ApoER2/LRP8 Function and Localization
- Low-density lipoprotein receptor-related protein 8 (LRP8), also known as Apolipoprotein E receptor 2 (ApoER2), is encoded by the LRP8 gene and undergoes alternative splicing to generate multiple isoforms.
- LRP8 is a cell surface receptor for Reelin (RELN) and apolipoprotein E-containing ligands, transmitting signals by binding DAB1 to regulate neuronal microtubule function and layer formation during brain development. Related references: PMID:12899622, PMID:12950167, PMID:20223215
- The receptor binds dimers of beta 2-glycoprotein I, potentially suppressing platelet aggregation in the vasculature. Related references: PMID:12807892
- LRP8 is highly expressed in the initial segment of the epididymis, where it regulates proteins essential for sperm maturation.
- It acts as a receptor for Semliki Forest virus and tick-borne encephalitis virus, mediating viral cell attachment and internalization.
- Subcellularly, LRP8 localizes to the cell membrane and can be secreted; it also functions as an endocytic receptor.
- Predominantly expressed in brain and placenta, with additional expression in platelets and megakaryocytic cells, but absent from the liver.
- LRP8 contains EGF-like domains, calcium-binding sites, disulfide bonds, and undergoes post-translational modifications including glycosylation, phosphorylation, and ubiquitin conjugation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal peptide (amino acids 900–C-terminus); therefore, the antibody may recognize full-length and C-terminal isoforms, but consider potential epitope masking in cell-based assays.
- For Western blotting (WB), the predicted molecular weight is ~106 kDa; use an appropriate gel percentage (e.g., 8% acrylamide) and validate using human or rat tissue lysates known to express LRP8, such as brain or placenta.
- In immunofluorescence on paraffin sections (IF-P) and immunohistochemistry on paraffin sections (IHC-P), antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA buffer pH 9.0) should be optimized for tissue types expressing LRP8.
- For ELISA, test paired antibody compatibility if used in sandwich assays, as the polyclonal nature may provide high avidity but could cross-react with related receptors like VLDLR.
- Since LRP8 is expressed in the brain and reproductive tissues, include appropriate positive controls (e.g., mouse or rat brain lysates) and negative controls (e.g., liver tissue, which lacks expression) to confirm specificity.
- Given the role in viral entry, the antibody can be explored in infection studies, but confirm cross-reactivity with viral proteins or infected cell lines.
