Biological Background: APOE Function and Localization
-
APOE is an apolipoprotein that functions primarily in lipoprotein-mediated lipid transport between organs via plasma and interstitial fluids. It is a core component of plasma lipoproteins and is involved in their production, conversion, and clearance.

APOE associates with chylomicrons, VLDL, and IDL, and preferentially binds HDL. It interacts with multiple cellular receptors (LDLR, LRP1, LRP2, LRP8, VLDLR) to mediate lipoprotein uptake. APOE also binds heparin and heparan-sulfate proteoglycans, facilitating lipoprotein capture.

In reverse cholesterol transport, APOE cooperates with ABCA1 to generate HDL particles and mediates hepatic HDL uptake, thereby regulating cholesterol homeostasis.
Related references: PMID:14754908, PMID:23620513, PMID:9395455 -
APOE is critical for lipid transport in the central nervous system, supporting neuron survival and sprouting.
Related references: PMID:25173806, PMID:8939961 -
It participates in innate and adaptive immunity, and binds the immune cell receptor LILRB4.
Related references: PMID:30333625 -
During HCV infection, APOE interacts with the E2 envelope glycoprotein, promoting viral attachment to host cells and contributing to viral particle assembly and infectivity.
Related references: PMID:25122793, PMID:29695434 -
APOE is secreted and found in plasma, interstitial fluid, and lymph. Mainly produced by hepatocytes, it is also expressed significantly in brain (astrocytes, neurons), peripheral nervous system, adrenal gland, testis, ovary, kidney, spleen, adipose tissue, and macrophages. Subcellularly, it localizes to the extracellular space, matrix, and endosomes/multivesicular bodies.
Related references: PMID:25173806, PMID:10027417, PMID:3115992 -
APOE is a glycoprotein and phosphoprotein, subject to glycation. Variants are implicated in Alzheimer's disease, amyloidosis, and hyperlipidemia. (From UniProt keywords: Alzheimer disease, Amyloidosis, Disease variant)
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 41-307, which encompasses the receptor-binding domain and lipid-binding regions; the polyclonal nature may provide recognition of multiple epitopes, enhancing detection sensitivity.
- For Western blot, expect a band around 36 kDa. Validated sample types include human, mouse, and rat tissue or cell lysates; appropriate loading controls should be included.
- In IHC-P and IF/ICC, antigen retrieval and permeabilization steps may be required; typical localization patterns include cytoplasmic and extracellular staining, consistent with secreted and endosomal pools.
- For ELISA applications, direct coating of recombinant APOE or capture-sandwich formats can be considered; cross-reactivity with mouse and rat APOE makes the antibody suitable for preclinical models.
- As with any polyclonal reagent, lot-to-lot consistency should be monitored; it is advisable to validate the antibody in the specific experimental system and application.
