Biological Background: APOBEC3G Function and Localization
- APOBEC3G (A3G) is a DNA dC->dU-editing enzyme belonging to the APOBEC3 family of cytidine deaminases. It exists in multiple isoforms due to alternative splicing.
- It acts as a potent restriction factor against Vif-deficient HIV-1 by deaminating cytidine to uridine in single-stranded viral DNA, causing G-to-A hypermutations in the proviral genome. Related references: PMID:12808465, PMID:12808466, PMID:12809610
- The antiviral activity extends to other retroviruses including SIV, HBV, EIAV, XMRV, and SFV, and also inhibits LTR and non-LTR retrotransposon mobility. Related references: PMID:15031497, PMID:16378963, PMID:18448976, PMID:16527742
- A deamination-independent mechanism contributes to antiretroviral effects before proviral integration. Related references: PMID:12808465, PMID:12808466, PMID:12809610
- Subcellularly, APOBEC3G localizes to the cytoplasm, nucleus, and P-bodies, with its distribution influenced by cellular activation state.
- Tissue expression is observed in spleen, testes, ovary, peripheral blood leukocytes, and CD4+ lymphocytes. In resting CD4 T-cells and monocytes, it exists as a low molecular mass (LMM) form, switching to high molecular mass (HMM) complexes upon activation or differentiation, correlating with increased HIV-1 susceptibility.
- Keywords: Antiviral defense, innate immunity, metal-binding (zinc), phosphoprotein, and ubiquitylation highlight its multifaceted regulation and role in host-virus interactions.
Experimental Guidance and Technical Tips
- The immunogen spans the cytidine deaminase domain (aa 60-330), thus the antibody is likely to recognize both native and denatured APOBEC3G.
- For Western blot, expect a band at approximately 46kDa; include appropriate positive controls from human, mouse, or rat lysates.
- IHC-P may require optimization of antigen retrieval conditions; validate staining using tissue known to express APOBEC3G.
- Specificity can be confirmed by comparing signals in APOBEC3G knockout or knockdown samples versus wild-type.
