Biological Background: APOBEC3B Function and Localization

  • APOBEC3B (DNA dC->dU-editing enzyme APOBEC-3B), also known as phorbolin-1-related protein, is a member of the APOBEC3 family of cytidine deaminases.
  • Functions as a DNA deaminase that inhibits retrovirus replication and retrotransposon mobility through both deaminase-dependent and -independent mechanisms.
  • Converts cytosine to uracil in minus-sense single-stranded viral DNA, leading to G-to-A hypermutations in proviral DNA; it selectively targets single-stranded DNA and does not deaminate double-stranded DNA or RNA.
  • Exhibits antiviral activity against simian immunodeficiency virus (SIV), hepatitis B virus (HBV), and human T-cell leukemia virus type 1 (HTLV-1), and may inhibit the mobility of LTR and non-LTR retrotransposons.
  • Localizes predominantly to the nucleus.
