Biological Background: APOA5 Function and Localization
- APOA5 is a minor apolipoprotein mainly associated with HDL, VLDL, and chylomicrons, and is a critical regulator of plasma triglyceride levels.
- It stimulates lipoprotein lipase (LPL) activity through apo-CII and inhibits hepatic VLDL-triglyceride production, thereby modulating lipid metabolism.
- APOA5 binds heparin, which may facilitate its interactions with lipoproteins and cell surfaces.

- It poorly activates LCAT and does not promote cholesterol efflux from macrophages, indicating a specialized role distinct from other apolipoproteins.
- The protein is secreted and localizes to early and late endosomes, as well as the trans-Golgi network.
- Expression is primarily restricted to the liver and plasma.
- Post-translational modifications include phosphorylation, and variants in APOA5 are associated with diseases affecting lipid homeostasis.
- As a member of the apolipoprotein family, APOA5 contains coiled-coil domains and is involved in lipid transport and VLDL assembly.
Experimental Guidance and Technical Tips
- For Western blotting, liver or plasma samples from human or mouse are suitable starting materials; optimization of antibody dilution and denaturing/reducing conditions is recommended.
- The immunogen corresponds to amino acids 207–366 of human APOA5, which may influence epitope recognition; consider using the recombinant protein as a positive control.
- In ELISA applications, direct or sandwich formats can be explored; pairing with a capture antibody may require empirical testing.
- Cross-reactivity with mouse APOA5 is reported, but sequence alignment in the target species should be verified for other applications.
