- APIP, also known as APAF1-interacting protein (hAPIP), is a methylthioribulose-1-phosphate dehydratase enzyme encoded by the APIP gene in humans.
- It catalyzes the dehydration of methylthioribulose-1-phosphate (MTRu-1-P) into 2,3-diketo-5-methylthiopentyl-1-phosphate (DK-MTP-1-P), a key step in the methionine salvage pathway.
- The methionine salvage pathway is crucial for cancer cell metabolism, apoptosis regulation, microbial proliferation, and inflammation.
- APIP inhibits CASP1-related inflammatory response (pyroptosis), the CASP9-dependent apoptotic pathway, and cytochrome c-dependent, APAF1-mediated cell death.
- APIP is primarily localized in the cytoplasm.
- Two isoforms exist: Isoform 1 is ubiquitously expressed, while isoform 2 shows lower expression and is detected in heart, brain, pancreas, liver, placenta, skeletal muscle, and kidney.
