Biological Background: APEH Function and Localization
- Official full name: Acylamino-acid-releasing enzyme, also known as acyl-peptide hydrolase (APH), acylaminoacyl-peptidase, and oxidized protein hydrolase (OPH).
- Functions as an aminopeptidase with exopeptidase activity, specifically hydrolyzing the N-terminal peptide bond of N-acetylated peptides to generate an N-acetylated amino acid and a peptide with a free N-terminus; preferentially cleaves Ac-Ala, Ac-Met, and Ac-Ser. - Exhibits endopeptidase activity involved in the degradation of oxidized and glycated proteins, contributing to cellular quality control of damaged proteins. Related references: PMID:10719179
- May act sequentially with aminoacylase-1 (ACY1) in the degradation of N-acylated peptides: APEH first removes N-acylamino acids from N-acylated peptides, then ACY1 further hydrolyzes the N-acylamino acid into free amino acid and a carboxylate.
- Subcellular localization is predominantly cytoplasmic.
- Expressed in erythrocytes at the protein level, suggesting a role in red blood cell function.
- Post-translational modifications include acetylation and phosphorylation, as identified by proteomics.

- The immunogen corresponds to amino acids 1-260 of human APEH; for Western blot, the antibody is expected to detect a band at approximately 81 kDa. Use appropriate reducing conditions and loading controls.
- For immunoprecipitation, validate the antibody’s performance with your target cell/tissue lysate and consider optimizing antibody-to-lysate ratios.
- In ELISA, the antibody may recognize both native and denatured APEH; empirical determination of coating conditions and concentrations is recommended.
