Biological Background: APEH Function and Localization

  • Official full name: Acylamino-acid-releasing enzyme, also known as acyl-peptide hydrolase (APH), acylaminoacyl-peptidase, and oxidized protein hydrolase (OPH).
  • Functions as an aminopeptidase with exopeptidase activity, specifically hydrolyzing the N-terminal peptide bond of N-acetylated peptides to generate an N-acetylated amino acid and a peptide with a free N-terminus; preferentially cleaves Ac-Ala, Ac-Met, and Ac-Ser.   • Exhibits endopeptidase activity involved in the degradation of oxidized and glycated proteins, contributing to cellular quality control of damaged proteins. Related references: PMID:10719179
  • May act sequentially with aminoacylase-1 (ACY1) in the degradation of N-acylated peptides: APEH first removes N-acylamino acids from N-acylated peptides, then ACY1 further hydrolyzes the N-acylamino acid into free amino acid and a carboxylate.
  • Subcellular localization is predominantly cytoplasmic.
  • Expressed in erythrocytes at the protein level, suggesting a role in red blood cell function.
