Biological Background: APC Function and Localization
- APC is a tumor suppressor protein encoded by the APC gene, also known as deleted in polyposis 2.5 (DP2.5). It acts as a key negative regulator of the Wnt signaling pathway by promoting the rapid degradation of beta-catenin (CTNNB1).
- APC associates with both microtubules and actin filaments, mediating the organization of F-actin into ordered bundles. It also functions downstream of Rho GTPases and DIAPH1 to selectively stabilize microtubules. Related references: PMID:17293347
- The protein is required for the localization of MACF1 to the cell membrane, which is critical for microtubule stabilization at the cell cortex.
- APC plays a role in hepatocyte growth factor (HGF)-induced cell migration and is necessary for MMP9 up-regulation via the JNK signaling pathway in colorectal tumor cells.
- APC is localized at cell junctions (adherens junctions), the cytoplasm, cytoskeleton, cell projections (lamellipodia and ruffle membranes), and the cell membrane.
- APC is expressed in a variety of tissues including brain, small intestine, colon, thymus, skeletal muscle, heart, prostate, lung, spleen, ovary, testis, kidney, placenta, blood, and liver. Isoform 1A is strongly expressed in brain, while isoform 1B is predominant in other tissues.
- The protein undergoes phosphorylation, acetylation, and ubiquitination, and its activity is correlated with its phosphorylation state. It contains coiled-coil domains and ARM repeats.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 2600-2760 of human APC (recombinant fusion protein). For Western blot, a band at approximately 312 kDa is anticipated; consider using a low-percentage gel for large proteins and appropriate transfer conditions.
- In immunofluorescence (IF), expect staining at adherens junctions, lamellipodia, and cytoskeletal structures, consistent with reported subcellular localization. Validate in relevant cell lines.
- For immunohistochemistry (IHC-P), test on tissue sections with known APC expression, such as colon or brain, with appropriate antigen retrieval.
- In ELISA, this antibody may be used as capture or detection antibody; titrate for optimal performance in your assay system.
