Biological Background: APC Function and Localization
- APC (Adenomatous polyposis coli protein) is a 312 kDa tumor suppressor that functions as a negative regulator of the Wnt signaling pathway by promoting rapid degradation of CTNNB1 (β-catenin).
- It plays a role in cell migration, being activated by hepatocyte growth factor (HGF) and required for MMP9 up-regulation via the JNK signaling pathway in colorectal tumor cells.
- APC associates with both microtubules and actin filaments, mediating the organization of F-actin into ordered bundles and stabilizing microtubules, particularly at the cell cortex, through interactions with ERBB2, Rho GTPases, and DIAPH1. Related references: PMID:17293347
- The protein localizes to adherens junctions, cytoplasm, cytoskeleton, lamellipodia, ruffle membranes, and the cell membrane, reflecting its dynamic roles in cell adhesion and motility.
- Multiple alternative isoforms exist; isoform 1A is very strongly expressed in the brain, while isoform 1B is the predominant form in most other tissues, including the gastrointestinal tract and blood.
- APC contains several functional domains and motifs, including coiled-coil regions, armadillo repeats, and β-catenin binding sites, and is subject to phosphorylation, acetylation, and ubiquitination, which modulate its tumor suppressor activities.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning amino acids 2600–2760 of human APC, located in the C‑terminal region. This design may contribute to recognition of multiple isoforms and species.
- For western blotting, the predicted molecular weight is approximately 312 kDa; consider using a low‑percentage (e.g., 6–8%) SDS‑PAGE gel and extended transfer times for optimal resolution.
- In immunohistochemistry (IHC‑P) and immunofluorescence (IF/ICC), signal specificity should be confirmed using appropriate positive and negative control tissues or cells that express APC.
- ELISA can be employed for detection; optimal antibody dilution and blocking conditions must be determined empirically for each assay format.
- Cross‑reactivity with human, mouse, and rat APC is reported; when working with other species, validate reactivity in the relevant sample system.
