Biological Background: AP2M1 Function and Localization
- AP2M1 encodes the mu subunit of the adaptor protein complex 2 (AP-2), also known as AP-2 mu chain, clathrin coat assembly protein AP50, or adaptin-mu2.
- AP-2 functions in clathrin-dependent endocytosis by linking clathrin to the cell membrane and selecting cargo via Y-X-X-Φ and [ED]-X-X-X-L-[LI] endocytosis signal motifs. Related references: PMID:12694563 PMID:12952941 PMID:14745134
- The mu subunit specifically binds to Y-X-X-Φ motifs on transmembrane cargo proteins, with binding accessibility regulated by phosphorylation at Thr-156 following clathrin assembly and AAK1 kinase activation. Related references: PMID:11877457
- AP2M1 also contributes to non-clathrin endocytic pathways, such as ARF6-regulated trafficking, and mediates endocytosis of ADAM10 during hippocampal long-term potentiation. Related references: PMID:19033387 PMID:23676497
- The protein is localized to the cell membrane and clathrin-coated pits, consistent with its role in endocytosis.
- AP2M1 is expressed in the brain at the protein level, and mutations in this gene have been linked to intellectual disability and epilepsy (Disease variant).
- Keywords associated with AP2M1 include endocytosis, lipid-binding, phosphoprotein, and protein transport, highlighting its multifunctional role in membrane traffic.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 134–433 of human AP2M1, spanning a region that includes the cargo-binding domain; consider validating antibody specificity in the relevant sample system.
- For Western blot (WB), a band at approximately 50 kDa is expected; optimal antibody dilution and exposure times should be determined empirically.
- In ELISA applications, the antibody may be used for antigen capture or detection; performance should be assessed with the specific assay format and sample matrix.
- The antibody is validated for human samples; cross-reactivity with other species has not been tested, so validation is recommended if used with samples from other organisms.
