Biological Background: Annexin A7 Function and Localization
- Annexin A7 (also known as annexin VII, synexin) is a member of the annexin family of calcium-dependent phospholipid-binding proteins.
- The protein contains four annexin repeats, which mediate its calcium-dependent interaction with acidic phospholipids in cell membranes.
- ANXA7 plays a critical role in membrane fusion events, including the regulation of exocytosis in various cell types.
- Two major isoforms exist due to alternative splicing: isoform 1 is predominantly expressed in brain, heart, and skeletal muscle, while isoform 2 is enriched in liver, lung, kidney, spleen, fibroblasts, and placenta.
- Post-translational modifications, including acetylation, may regulate its function and subcellular localization.
- ANXA7 has been implicated in diverse cellular processes such as calcium signaling, membrane trafficking, and apoptosis.
- The protein exhibits a molecular weight of approximately 53 kDa (as per product specifications) or 52.7 kDa (UniProt), varying with isoform and experimental conditions.
Experimental Guidance and Technical Tips
- The immunogen, corresponding to amino acids 1-250 of human ANXA7, targets the N-terminal region, which may confer specificity for ANXA7 over other annexin family members.
- For Western blotting (WB), the antibody is expected to detect a band around 53 kDa; however, the presence of multiple isoforms or post-translational modifications could lead to additional bands, so validate in the relevant sample system.
- In immunofluorescence/immunocytochemistry (IF/ICC), as ANXA7 is primarily localized to the cytoplasm and can translocate to membranes upon calcium activation, optimize fixation and permeabilization conditions to maintain epitope integrity.
- For ELISA applications, the antibody can be used for quantitative detection of ANXA7; consider pairing with a detection antibody against a different epitope to develop a sensitive sandwich ELISA.
- Given the reported cross-reactivity with human, mouse, and rat, the antibody is suitable for studies in these species, but verification in other species is recommended if needed.
