Biological Background: ANXA13 Function and Localization

  • ANXA13 encodes annexin A13, a member of the annexin family of calcium-dependent phospholipid-binding proteins. Two isoforms are produced by alternative splicing: isoform A (canonical) and a variant with distinct calcium sensitivity.
  • Isoform A binds to membranes enriched in phosphatidylserine or phosphatidylglycerol in a calcium-dependent manner, with half-maximal binding at ~60 µM Ca²⁺. It does not associate with membranes lacking acidic phospholipids. Related references: PMID:27676605 PMID:30610115
  • The alternative isoform also requires calcium for membrane interaction but exhibits lower affinity, requiring ~320 µM Ca²⁺ for half-maximal binding.
  • Subcellularly, ANXA13 localizes to the apical cell membrane, cell membrane, and cytoplasmic vesicles, consistent with a role in membrane trafficking and organization.
  • ANXA13 is predominantly expressed in intestinal epithelial cells, with protein detected in colon and jejunum, supporting its alias "Intestine-specific annexin" (ISA).

  • The immunogen used is a recombinant protein covering amino acids 1-316 of human ANXA13 (full-length canonical sequence). This may enable recognition of both isoforms, but verification is advised.
  • For western blot (WB), the predicted molecular weight is ~35 kDa. Use whole-cell lysates or membrane fractions from intestinal cell lines (e.g., Caco-2, HT-29) or tissue homogenates (colon, jejunum).
  • For immunohistochemistry (IHC-P), antigen retrieval protocols and antibody dilution should be optimized in positive control tissues such as human colon or jejunum sections.
  • In ELISA applications, the antibody can be used for direct or indirect detection of recombinant or native ANXA13, with appropriate secondary antibodies and colorimetric/fluorescent substrates.
  • Cross-reactivity with mouse and rat ANXA13 is indicated; however, users should validate in the relevant sample system to confirm specificity.
