Biological Background: ANT2/SLC25A5 Function and Localization
- ANT2/SLC25A5, also known as ADP/ATP translocase 2, adenine nucleotide translocator 2, or solute carrier family 25 member 5, is a mitochondrial inner membrane protein belonging to the SLC25 family of mitochondrial carriers.
- It functions primarily as an ADP:ATP antiporter, exchanging cytosolic ADP for matrix ATP, thereby fueling cellular energy metabolism.
- SLC25A5 also exhibits uncoupling activity by facilitating proton transport, which reduces the efficiency of ATP synthesis and contributes to mitochondrial thermogenesis, particularly in tissues lacking UCP1.
- The protein is implicated in the opening of the mitochondrial permeability transition pore (mPTP), a non-specific high-conductance channel that triggers cell death. Related references: PMID:31883789
- Beyond its transport roles, SLC25A5 regulates mitophagy through interaction with TIMM44, stabilizing PINK1, and participates in chromosome segregation as part of the MMXD complex. Related references: PMID:20797633
- It localizes exclusively to the inner mitochondrial membrane, with the majority of the protein spanning the membrane via six transmembrane helices.
- Post-translational modifications include acetylation, methylation, and phosphorylation, which may regulate its activity and interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (amino acids 120–220) of human ANT2/SLC25A5; this region is likely well-conserved across indicated reactive species (human, mouse, rat), but sequence alignment should be verified for other species.
- For Western blotting, a band around 33 kDa is expected in human, mouse, and rat mitochondrial lysates. Inclusion of appropriate positive and negative controls is recommended.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), mitochondrial staining patterns should be observed; fixation and antigen retrieval conditions may need optimization for specific sample types.
- This antibody is also suitable for ELISA applications; consider standard indirect ELISA protocols and validate with recombinant or native protein.
- Since ANT2 is a multi-pass membrane protein, denaturing conditions and reducing agents are critical for consistent WB results.
