  • ANT1 (SLC25A4) is an ADP/ATP antiporter that catalyzes the exchange of mitochondrial matrix ATP for cytosolic ADP, a central step in cellular energy metabolism.

  • The protein operates via an alternating access mechanism, cycling between cytoplasmic-open and matrix-open states to alternately expose a single substrate-binding site to either side of the inner mitochondrial membrane.
  • ANT1 also mediates mitochondrial uncoupling by acting as a proton transporter, a process that diminishes ATP synthesis efficiency and contributes to thermogenesis; this activity requires free fatty acids as cofactors.
  • It participates in the mitochondrial permeability transition pore (mPTP), a non-specific pore implicated in cell death.
