Biological Background: ANT1/ANT2/ANT3/ANT4 Function and Localization
- ANT1 (SLC25A4) is an ADP/ATP antiporter that catalyzes the exchange of mitochondrial matrix ATP for cytosolic ADP, a central step in cellular energy metabolism.
Related references: PMID:21586654 PMID:27693233 PMID:23173940 - The protein operates via an alternating access mechanism, cycling between cytoplasmic-open and matrix-open states to alternately expose a single substrate-binding site to either side of the inner mitochondrial membrane.
- ANT1 also mediates mitochondrial uncoupling by acting as a proton transporter, a process that diminishes ATP synthesis efficiency and contributes to thermogenesis; this activity requires free fatty acids as cofactors.
- It participates in the mitochondrial permeability transition pore (mPTP), a non-specific pore implicated in cell death.
Related references: PMID:31883789 - Independently of its transport function, ANT1 regulates mitophagy by interacting with TIMM44, which inhibits the TIM23 translocase and stabilizes PINK1.
- The protein resides in the mitochondrial inner membrane and is subject to multiple post-translational modifications, including acetylation, methylation, phosphorylation, and S-nitrosylation.
- The antibody recognizes all four adenine nucleotide translocase isoforms (ANT1–4), which share conserved sequences in the C-terminal region.
Experimental Guidance and Technical Tips
- The polyclonal antibody was raised against a synthetic peptide from the C-terminus of human ANT1, a region highly conserved among ANT isoforms, which likely accounts for its broad reactivity with ANT1–4.
- For Western blot, expected bands are around 33 kDa (ANT1), 32 kDa (ANT2/3), and 35 kDa (ANT4); use appropriate loading controls and validate band identity with knockdown or knockout samples.
- In IHC-P, optimize antigen retrieval and antibody dilution on known positive control tissues (e.g., heart or skeletal muscle) before analyzing experimental samples.
- For ELISA, consider using the peptide immunogen as a positive coating antigen to verify antibody titer.
