Biological Background: ANP32A Function and Localization
- ANP32A, also known as LANP, pp32, and PHAPI, is a multifunctional protein involved in tumor suppression, apoptosis, cell cycle progression, and transcriptional regulation. Related references: PMID:10400610 PMID:11360127 PMID:16341127
- It promotes apoptosis by facilitating caspase-9 activation and apoptosome formation. Related references: PMID:18439902
- As part of the INHAT (inhibitor of histone acetyltransferases) complex, ANP32A inhibits the histone-acetyltransferase activity of EP300/CREBBP and associated factors through histone masking. Related references: PMID:11830591
- It preferentially binds unmodified histone H3, sterically inhibiting its acetylation and phosphorylation, thereby contributing to cell growth inhibition. Related references: PMID:16341127
- ANP32A regulates mRNA nuclear-to-cytoplasmic translocation and stability by associating with the RNA-binding protein ELAVL1. Related references: PMID:18180367
- It also functions as an inhibitor of protein phosphatase 2A and participates in E4F1-mediated transcriptional repression. Related references: PMID:15642345 PMID:17557114
- During microbial infection, ANP32A is essential for influenza A, B, and C viral genome replication by mediating assembly of the viral replicase. Related references: PMID:30666459 PMID:32694517 PMID:33045004 PMID:33208942 PMID:21159877
- Subcellular localization includes the nucleus, cytoplasm, and endoplasmic reticulum. It is ubiquitously expressed, with highest levels in kidney and skeletal muscle, and moderate in brain, placenta, and pancreas.
Experimental Guidance and Technical Tips
- For Western blot (WB), the predicted molecular weight is ~29 kDa; however, post-translational modifications or oligomerization may result in additional bands. Consider using appropriate positive controls.
- For immunohistochemistry (IHC-P), validate staining with tissues showing high ANP32A expression such as kidney or skeletal muscle. Antigen retrieval conditions may require optimization.
- For ELISA applications, test coating and detection conditions using known concentrations of recombinant protein. The polyclonal nature may introduce variability; include negative controls.
- As the immunogen corresponds to the N-terminal 1–170 amino acids, the antibody may recognize full-length and truncated isoforms. Cross-reactivity with mouse and rat is reported; however, verify in the specific experimental system.
