Biological Background: ANO1 Function and Localization
- ANO1 (Anoctamin-1) is a member of the anoctamin family, also designated DOG1, TMEM16A, ORAOV2, or TAOS2. It is a 114 kDa multi-pass transmembrane protein with a conserved calcium-binding domain and predicted N-glycosylation sites.
- ANO1 functions as a calcium-activated chloride channel (CaCC) that mediates anion secretion across epithelia and contributes to smooth muscle contraction and pacemaker activity in gastrointestinal interstitial cells of Cajal. Related references: PMID:20056604, PMID:22178883, PMID:22946059
- It is required for CFTR activation by enhancing endoplasmic reticulum Ca²⁺ store release and supporting CFTR membrane expression. Related references: PMID:28963502
- ANO1 contributes to basal and ATP-dependent mucus secretion in airways and intestine, and its activity has been linked to interleukin- and CLCA1-induced MUC5AC mucin expression, although findings are contradictory. Related references: PMID:33026825, PMID:31732694
- The protein localizes to the apical cell membrane and presynaptic compartments in relevant tissues.
- ANO1 is broadly expressed, with higher levels in liver, skeletal muscle, and gastrointestinal muscles; it is also detected in nasal epithelia, kidney collecting ducts, and eccrine sweat glands. Related references: PMID:32487539, PMID:24913262, PMID:15215166, PMID:16906560, PMID:25220078
- Key post-translational modifications include phosphorylation, glycosylation, and disulfide bond formation; alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen spans residues 400–500 of human ANO1, an intracellular region between transmembrane domains. This design may enable recognition of native and denatured forms, supporting WB and ELISA applications.
- For immunofluorescence/ICC, consider using permeabilization steps to allow antibody access to the intracellular epitope.
- When performing western blotting, note the predicted molecular weight of ~114 kDa; validate band specificity using appropriate positive and negative lysates (e.g., human and mouse gastrointestinal or airway epithelial cells).
- Cross-reactivity with mouse is indicated; verify in your specific model system, as mouse ANO1 shares high sequence identity in the immunogen region.
- Optimal working dilutions must be determined experimentally for each application.
