Biological Background: Annexin A2 Function and Localization

  • Annexin A2 (also known as p36, lipocortin II, calpactin I heavy chain) is a member of the annexin family of calcium-dependent phospholipid-binding proteins.
  • It is a calcium-regulated membrane-binding protein that binds two calcium ions with high affinity; this binding is greatly enhanced by anionic phospholipids. May also be involved in the heat-stress response.
  • Annexin A2 inhibits PCSK9-enhanced LDLR degradation, likely by reducing PCSK9 protein levels through a translational mechanism and competing with LDLR for PCSK9 binding. Related references: PMID:18799458 PMID:22848640 PMID:24808179
  • It binds to endosomes damaged by phagocytosis of particulate wear debris and participates in endosomal membrane stabilization, thereby limiting NLRP3 inflammasome activation.
  • Required for endothelial cell surface plasmin generation and may support fibrinolytic surveillance and neoangiogenesis.
  • Secreted into the extracellular space and found in the extracellular matrix, basement membrane, and melanosomes. Post-translational modifications include acetylation, phosphorylation, isopeptide bond formation, and ubiquitination. The protein undergoes alternative splicing and has RNA-binding capability.

  • The immunogen is a recombinant protein covering the full-length human Annexin A2 (amino acids 1-339). This polyclonal antibody may recognize both linear and conformational epitopes; consider validating specificity in your target sample.
  • For Western blot (WB), an expected band around 39 kDa is observed. Due to post-translational modifications, slight variations in molecular weight may occur; validate using appropriate positive and negative controls.
  • For immunohistochemistry (IHC-P), test both paraffin-embedded and frozen sections and optimize antigen retrieval conditions.
  • For immunofluorescence (IF/ICC), fixation and permeabilization steps should be optimized for your cell type to ensure accessibility to intracellular antigens.
  • This antibody cross-reacts with mouse and rat Annexin A2, making it suitable for studies in these species. Species-specific validation is recommended.
