Biological Background: Annexin A1 Function and Localization
- Annexin A1, also known as Annexin I, Lipocortin I, Calpactin II, p35, and chromobindin-9, is a member of the annexin superfamily characterized by conserved calcium/phospholipid-binding repeats.
- It functions as an anti-inflammatory protein by mediating the effects of glucocorticoids and down-regulating the early phase of the inflammatory response (PubMed:8425544). Related references: PMID:8425544
- In adaptive immunity, Annexin A1 promotes the differentiation of T cells into Th1 cells while negatively regulating Th2 differentiation, thereby modulating T-cell activation and proliferation (PubMed:17008549). Related references: PMID:17008549
- The protein exhibits high-affinity Ca²⁺-dependent binding to phospholipid membranes and can bind up to eight Ca²⁺ ions, which is essential for its membrane association and function (PubMed:2532504; PubMed:8557678). Related references: PMID:2532504 PMID:8557678
- Annexin A1 signals through formyl peptide receptors (FPRs) to promote chemotaxis of granulocytes and monocytes, actin cytoskeleton rearrangement, cell polarization, and migration, and it plays a role in the resolution of inflammation and wound healing (PubMed:15187149; PubMed:22879591; PubMed:25664854). Related references: PMID:15187149 PMID:22879591 PMID:25664854
- It negatively regulates hormone exocytosis via FPR activation and actin reorganization (PubMed:19625660). Related references: PMID:19625660
- Subcellular localization is diverse, including the nucleus, cytoplasm, cell membrane, secretory vesicles, exosomes, and phagocytic cups, reflecting its multifunctional nature.
- Tissue expression is detected in resting neutrophils, peripheral blood T cells, placenta, liver, and extracellular vesicles from inflammatory bowel disease sera, consistent with its roles in immune and inflammatory processes.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 2–346, covering nearly the full-length human Annexin A1 protein; this design often yields antibodies that recognize both native and denatured forms, making them suitable for multiple applications.
- For Western Blot (WB) and Immunohistochemistry (IHC), we recommend titrating the antibody in the relevant sample matrix to determine the optimal working concentration.
- In Immunofluorescence/Immunocytochemistry (IF/ICC), fixation and permeabilization conditions may need optimization due to Annexin A1’s presence in multiple subcellular compartments.
- The antibody has been verified for immunoprecipitation (IP) and ELISA; consider using appropriate positive controls such as stimulated immune cells or purified protein.
- Cross-reactivity with mouse and rat indicates conserved epitopes, but users should validate in their specific model systems.
