Biological Background: ANKRD28 Function and Localization

  • ANKRD28, also known as Serine/threonine-protein phosphatase 6 regulatory ankyrin repeat subunit A, PITK, FAP79, CFAP79, PPP1R65, is a regulatory subunit of protein phosphatase 6 (PP6) that mediates substrate recognition and targeting.
  • It is involved in PP6-mediated dephosphorylation of NFKBIE (IκBε), opposing its TNF-induced degradation and thus modulating NF-κB signaling.
  • ANKRD28 selectively inhibits the phosphatase activity of protein phosphatase 1 catalytic subunit (PPP1C) and targets PPP1C to modulate heterogeneous nuclear ribonucleoprotein K (HNRPK) phosphorylation.
  • It participates in PP6-dependent dephosphorylation of MOB1, promoting focal adhesion assembly during cell migration. Related references: PMID:35512830
  • The protein contains ankyrin repeat domains and is localized in the nucleoplasm, cytosol, and at lamellipodia, consistent with roles in both nuclear and cytosolic signaling.
  • Post-translational modifications include phosphorylation and ubiquitin conjugation, and the gene is subject to alternative promoter usage and splicing.

Experimental Guidance and Technical Tips

  • For Western blot (WB), consider testing a dilution range of 1:500 to 1:2000 and validate detection of endogenous ANKRD28 in relevant cell lysates (e.g., HeLa, HEK293). The predicted molecular weight is 113 kDa, but observe for possible post-translational modifications or splice isoforms.
  • For ELISA, pair this antibody with a matched detection reagent and titrate from 0.1–1.0 µg/mL; include appropriate positive and negative controls.
  • The immunogen spans the N-terminal 300 amino acids; so the antibody may recognize multiple isoforms. Verify reactivity in your specific experimental system.
  • This antibody cross-reacts with human and mouse; when using mouse samples, perform a species-specific validation to confirm performance.
