Biological Background: ANKRD1 Function and Localization
- ANKRD1, also known as cardiac ankyrin repeat protein (CARP) and cytokine-inducible gene C-193 protein, is characterized by ankyrin repeat and coiled-coil domains.
- It may function in endothelial cell activation and as a nuclear transcription factor that negatively regulates cardiac gene expression.
- Induction of ANKRD1 has been correlated with apoptotic cell death in hepatoma cells.
- Localized primarily to the nucleus.
- Mainly expressed in activated vascular endothelial cells, with lower expression in hepatoma cells.
- Contains ankyrin repeat motifs and a coiled-coil region, features typical of transcriptional regulators.
Experimental Guidance and Technical Tips
- The immunogen sequence covers the N-terminal 130 amino acids, a region that includes predicted ankyrin repeat motifs; thus, the antibody may detect full-length ANKRD1 (~36 kDa) on Western blots.
- For Western blot, consider preparing nuclear extracts to enrich the target protein, given its nuclear localization.
- In ELISA, optimize coating antigen concentrations and antibody dilutions using recombinant ANKRD1 or cell lysates as positive controls.
- Validate cross-reactivity in mouse and rat samples (e.g., endothelial or hepatoma cell lines) to confirm binding.
- Include appropriate negative controls (e.g., non-transfected cells or tissues with low ANKRD1 expression) to assess specificity.
