Biological Background: ANKLE2 Function and Localization
- ANKLE2 (Ankyrin repeat and LEM domain-containing protein 2), also known as Lem4 or LEMD4, is a transmembrane protein that belongs to the LEM domain family.
- It plays a critical role in mitotic nuclear envelope reassembly by promoting dephosphorylation of barrier-to-autointegration factor (BAF/BANF1) during mitotic exit. Related references: PMID:22770216
- ANKLE2 coordinates control of BAF dephosphorylation by inhibiting VRK1 kinase and enhancing dephosphorylation by protein phosphatase 2A (PP2A), facilitating nuclear envelope formation. Related references: PMID:22770216
- It may regulate nuclear localization of VRK1 in non-dividing cells, though its precise role as a PP2A regulatory subunit remains unclear. Related references: PMID:31735666
- ANKLE2 is essential for normal brain development; mutations are associated with primary microcephaly (MCPH16). Related references: PMID:25259927
- It localizes to the endoplasmic reticulum membrane and contains a signal-anchor, ANK repeats, and a transmembrane helix.
- ANKLE2 is a phosphoprotein, with alternative splicing producing multiple isoforms, and it is implicated in cell cycle and division processes.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein corresponding to amino acids 350–450 of human ANKLE2 (NP_055929.1). Consider this region when designing epitope mapping or competitive binding assays.
- This antibody is applicable to Western blotting (WB), immunohistochemistry on paraffin-embedded sections (IHC-P), and ELISA. Validate and optimize dilution factors in the relevant sample system.
- Predicted molecular weight of ANKLE2 is 104 kDa; confirm protein migration under your experimental conditions and account for potential post-translational modifications.
- Reported cross-reactivity includes human, mouse, and rat. Verify reactivity in your specific tissue or cell line to ensure suitability.
- For IHC-P, antigen retrieval and blocking conditions may need optimization based on tissue type and fixation.
