Biological Background: ANKHD1 Function and Localization

• ANKHD1 (Ankyrin repeat and KH domain-containing protein 1), also known as MASK, MASK1, VBARP, or PP2500, is a large scaffolding protein containing ankyrin repeats and a KH domain.
• It may function as a scaffolding protein associated with abnormal phenotypes in leukemia cells.
• Isoform 2 may have antiapoptotic effects by regulating caspases, protecting cells during normal survival.
• The protein localizes to the cytoplasm.
• It is ubiquitously expressed, with high levels in cervix, spleen, and brain; hematopoietic cells show increased expression in leukemia.
• Isoform 2 shows high expression in spleen but almost none in muscle and brain.
• Post-translational modifications include acetylation and phosphorylation; it also contains an RNA-binding KH domain.
