Biological Background: ANKH Function and Localization
- ANKH is a transmembrane protein that functions as an ATP and NTP transporter, primarily localized to the cell membrane.
- By exporting NTPs, ANKH provides substrates for ENPP1 to generate extracellular inorganic pyrophosphate (PPi), a key calcification inhibitor, thereby preventing abnormal joint mineralization. Related references: PMID:20943778, PMID:32639996, PMID:35147247
- ANKH also mediates the release of TCA cycle intermediates (citrate, succinate, malate) to the extracellular space, where citrate supports osteogenic differentiation and bone strength. Related references: PMID:32639996
- The protein is predominantly expressed in osteoblasts from mandibular and iliac bone, consistent with its role in bone mineralization.
- Alternative splicing produces multiple isoforms, and genetic variants in ANKH are linked to craniometaphyseal dysplasia, chondrocalcinosis, and deafness (UniProt keywords: Alternative splicing, Disease variant, Deafness).
Experimental Guidance and Technical Tips
- The immunogen targets a central region (aa 210-320) of human ANKH, which may recognize multiple isoforms depending on epitope conservation.
- For Western blot analysis, consider using positive control lysates from osteoblast cell lines (e.g., human osteosarcoma) given the tissue expression profile.
- For ELISA applications, verify antibody reactivity with recombinant ANKH protein and consider pairing with a detection antibody to improve sensitivity.
- The predicted molecular weight is 54 kDa; post-translational modifications may cause migration shifts.
- Cross-reactivity is reported for human; validate in other species if required for your experimental model.
