Biological Background: ANGPTL3 Function and Localization

  • ANGPTL3, also known as angiopoietin-5, is a member of the angiopoietin-like protein family encoded by the ANGPTL3 gene in humans.
  • It acts as a liver-derived hepatokine with a central role in the regulation of systemic lipid and glucose metabolism. Related references: PMID:11788823, PMID:12909640, PMID:23661675
  • ANGPTL3 significantly elevates plasma triglyceride levels by inhibiting lipoprotein lipase (LPL) activity through the recruitment of proprotein convertases PCSK6 and FURIN, which cleave and release LPL from the cell surface; this function is mediated by the N-terminal domain and does not require ANGPTL3 cleavage.   • It also inhibits endothelial lipase, leading to increased plasma levels of high-density lipoprotein (HDL) cholesterol and phospholipids. Related references: PMID:17110602, PMID:19028676
  • ANGPTL3 is involved in angiogenesis: it binds to endothelial cells via integrin alpha-V/beta-3, activates FAK, MAPK, and Akt signaling pathways, and promotes cell adhesion and migration.
