Biological Background: Angiopoietin-2 Function and Localization

  • ANGPT2, also known as ANG2, is a secreted glycoprotein that serves as a context-dependent regulator of angiogenesis, binding to the TIE2 receptor.
  • It competes with ANGPT1 for TIE2 binding, modulating downstream signaling; in the absence of VEGF, ANGPT2 promotes vascular regression by inducing endothelial cell apoptosis, while in the presence of VEGF it facilitates endothelial migration and proliferation, acting as a permissive angiogenic signal. Related references: PMID:15284220 PMID:19116766 PMID:19223473
  • ANGPT2 is also involved in the regulation of lymphangiogenesis. Related references: PMID:32908006
  • It is a secreted protein, synthesized with a signal peptide, and is found in the extracellular space.
  • The protein is post-translationally modified with glycosylation and disulfide bonds, and its structure includes a coiled-coil domain; alternative splicing generates multiple isoforms.
  • UniProt keywords include Angiogenesis, Developmental protein, Disease variant, Glycoprotein, Secreted, and 3D-structure, underscoring its role in vascular development and pathology.
  • The predicted molecular weight is approximately 57 kDa.

Experimental Guidance and Technical Tips

  • For Western blot (WB), run samples under reducing conditions and validate that the primary band appears near 57 kDa; additional bands may correspond to glycosylated variants or isoforms.
  • In immunofluorescence (IF/ICC), consider that ANGPT2 is primarily secreted; intracellular staining may require permeabilization and inclusion of secretion inhibitors (e.g., brefeldin A) to visualize the intracellular pool.
  • For ELISA, this polyclonal antibody may be used either as a capture or detection reagent after appropriate titration and pairing validation.
  • Given cross-reactivity with mouse and rat, validate the antibody in your specific model system and optimize blocking/dilution conditions to minimize species-specific background.
  • The immunogen covers amino acids 197–496, which is the central portion of the protein; it is expected to recognize all major isoforms, but confirm reactivity for your target isoform if isoform-specific detection is required.
