Biological Background: Angiogenin (ANG) Function and Localization
- Angiogenin (ANG), also known as ribonuclease 5 (RNase 5), is a member of the pancreatic ribonuclease superfamily with dual roles in cell proliferation and ribonuclease activity.
- It can either promote or restrict cell proliferation depending on the cellular context, acting as a secreted ribonuclease that is endocytosed via the PLXNB2 receptor. Related references: PMID:12051708, PMID:1400510, PMID:19332886
- Under stress conditions, ANG localizes to the cytoplasm and promotes stress granule assembly by cleaving specific tRNAs (e.g., tRNA(Ala), tRNA(Glu)) into tRNA-derived stress-induced fragments (tiRNAs), leading to translation repression and enhanced cell survival. Related references: PMID:1400510, PMID:19332886, PMID:20129916, PMID:31582561
- In growth conditions, ANG translocates to the nucleus and stimulates ribosomal RNA (rRNA) transcription, including 45S rRNA, thereby promoting cell growth, proliferation, and angiogenesis. Related references: PMID:12051708, PMID:15735021, PMID:27518564, PMID:19354288, PMID:4074709, PMID:8448182
- ANG is predominantly expressed in the liver, with additional detection in endothelial cells and spinal cord neurons, and is implicated in hematopoietic stem cell maintenance and intestinal epithelial barrier integrity. Related references: PMID:2440105, PMID:17886298, PMID:27518564, PMID:32510170
- Subcellular localization includes secretion, nucleus (nucleolus), cytoplasm, and stress granules; ANG is associated with keywords such as amyotrophic lateral sclerosis, angiogenesis, and stress response.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 25–147 of mature angiogenin, a region conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blot, the predicted molecular weight is 17 kDa; choose an appropriate gel percentage and transfer conditions, and validate with a positive control (e.g., liver lysate).
- In IF/ICC, consider that angiogenin can localize to cytoplasm, nucleus, or be secreted; include permeabilization and co-staining with organelle markers as needed.
- ELISA can be utilized to detect secreted angiogenin in serum or culture supernatants; optimize coating and detection conditions empirically.
