Biological Background: ANAPC5 Function and Localization

  • ANAPC5 (Anaphase-promoting complex subunit 5), also known as cyclosome subunit 5, is a core subunit of the anaphase-promoting complex/cyclosome (APC/C), a multi-subunit E3 ubiquitin ligase.
  • The APC/C mediates ubiquitination and degradation of target proteins, primarily forming 'Lys-11'-linked polyubiquitin chains and, to a lesser extent, 'Lys-48'- and 'Lys-63'-linked chains (PubMed:18485873).
    Related references: PMID:18485873
  • APC/C catalyzes the assembly of branched ubiquitin chains, such as 'Lys-11'/'Lys-48'-linked branches, on substrates to modulate protein turnover (PubMed:29033132).
    Related references: PMID:29033132
  • During the metaphase-to-anaphase transition, APC/C is activated by CDC20, triggering the degradation of cyclin B and securin to promote mitotic exit (PubMed:27120157, PubMed:27509861). The mitotic checkpoint complex (MCC) inhibits APC/C activity to delay chromosome segregation until all chromosomes are properly attached (PubMed:27509861).
    Related references: PMID:27120157, PMID:27509861
  • ANAPC5 localizes to the nucleus, cytoplasm, and mitotic spindle during cell division, reflecting its roles in both interphase and mitotic regulation.
  • ANAPC5 contains multiple tetratricopeptide repeat (TPR) domains, is subject to alternative splicing and phosphorylation, and belongs to the ubiquitin conjugation pathway, as identified in high-throughput proteomic analyses.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1–250 (N-terminal region) of human ANAPC5, so consider epitope mapping if targeting C-terminal isoforms.
  • For Western blot, the expected molecular weight is 85 kDa; validate with knockout (KO) controls to ensure specificity.
  • For immunohistochemistry (IHC-P), optimize antigen retrieval protocols as the antibody recognizes both native and denatured protein.
  • For immunoprecipitation (IP), include appropriate isotype controls and pre-clearing steps to reduce non-specific binding.
