Biological Background: AMPK alpha-1 Function and Localization
- AMPK alpha-1 is the catalytic subunit of the heterotrimeric AMP-activated protein kinase (AMPK), a central energy sensor that is activated by rising AMP/ATP ratios. It phosphorylates numerous downstream targets to restore energy balance by stimulating catabolic pathways and inhibiting anabolic processes.
Related references: PMID:17307971, PMID:17712357, PMID:24563466 - In lipid metabolism, it directly phosphorylates and inactivates acetyl-CoA carboxylase (ACACA) and HMG-CoA reductase (HMGCR) to suppress fatty acid and cholesterol synthesis. It also promotes lipolysis by phosphorylating choline kinase alpha (CHKA) on lipid droplets.
Related references: PMID:34077757 - The kinase stimulates glucose uptake in muscle by enhancing SLC2A4/GLUT4 translocation to the plasma membrane, and modulates glycolysis through PFKFB2 and PFKFB3 phosphorylation.
- AMPK alpha-1 acts as a transcriptional regulator by phosphorylating factors such as CRTC2, FOXO3, p53/TP53, and histone H2B, thereby coupling energy status to adaptive gene expression programs.
Related references: PMID:11518699, PMID:11554766, PMID:15866171 - Under nutrient limitation, it suppresses mTORC1 signaling by phosphorylating TSC2 and RPTOR, and initiates autophagy by activating ULK1. It also phosphorylates WDR24 and FNIP1 to tune mTORC1 activity in response to glucose and energetic stress.
Related references: PMID:14651849, PMID:18439900, PMID:20160076, PMID:36732624, PMID:37079666, PMID:21205641, PMID:28561066 - The protein localizes to the cytoplasm, nucleus, and late endosomes, consistent with its diverse functions. It also regulates cellular polarity and phosphorylates a broad set of substrates including CFTR, EEF2K, CASP6, and ALDH7A1.

Experimental Guidance and Technical Tips
- The immunogen covers a conserved N-terminal region (aa 5–280) shared by AMPKa1 and AMPKa2, supporting cross-reactivity with mouse and rat orthologs as indicated. Verify reactivity in your specific system before large-scale studies.
- For Western blot, expect bands at approximately 64 kDa (AMPKa1) and 65 kDa (AMPKa2). Optimize protein loading and blocking agents to resolve close bands; include a positive control lysate.
- In immunofluorescence, AMPK shows both cytoplasmic and nuclear staining. Test different fixation protocols (e.g., methanol vs. formaldehyde) and include appropriate secondary-only controls.
- For ELISA, consider using the immunogen peptide or whole-cell lysates to generate a standard curve. Titrate the antibody to determine optimal working concentrations.
