Biological Background: AMPKa1/AMPKa2 Function and Localization
- AMPK (AMP-activated protein kinase) is a heterotrimeric energy sensor composed of a catalytic α subunit (PRKAA1/α1 or PRKAA2/α2), a regulatory β subunit, and a γ subunit. The α1 subunit is a 64 kDa serine/threonine kinase critical for cellular energy homeostasis.
- In response to low ATP, AMPK activates catabolic pathways and inhibits anabolic processes such as protein, carbohydrate, and lipid biosynthesis, as well as cell growth and proliferation.
Related references:
PMID:17307971, PMID:17712357, PMID:24563466 - AMPK directly phosphorylates transcription regulators (e.g., CRTC2, FOXO3, histone H2B, HDAC5, p53) to modulate gene expression and chromatin structure in response to metabolic stress.
Related references:
PMID:11518699, PMID:11554766, PMID:15866171 - It negatively regulates mTORC1 signaling by phosphorylating TSC2, RPTOR, WDR24, and FNIP1, thereby inhibiting cell growth and promoting autophagy under nutrient-poor conditions.
Related references:
PMID:14651849, PMID:18439900, PMID:20160076, PMID:36732624, PMID:37079666 - AMPK promotes autophagy by directly phosphorylating and activating ULK1 and WDR45.
Related references:
PMID:21205641, PMID:28561066 - AMPK is localized in the cytoplasm and nucleus, and can associate with late endosomes, consistent with its roles in metabolic signaling and trafficking.
- Key post-translational features include phosphorylation (regulating its activity), nucleotide-binding (AMP/ATP), and glycosylation. The enzyme is involved in multiple pathways including lipid/sterol metabolism, autophagy, circadian rhythm, and Wnt signaling.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 13-300) spans a highly conserved region of PRKAA1/PRKAA2, explaining robust cross-reactivity with human, mouse, and rat samples.
- For Western blotting, endogenous AMPKα isoforms are detected as bands around 62–65 kDa; multiple bands may reflect differential splicing or phosphorylation states.
- IHC-P may require antigen retrieval (e.g., citrate buffer pH 6.0) and careful titration; inclusion of appropriate positive and negative tissue controls is recommended.
- ELISA applications can be established using recombinant protein or peptide standards; pair with appropriate secondary antibodies and optimize coating conditions.
