Biological Background: AMPKa1/AMPKa2 Function and Localization
- Energy sensor kinase: AMPKα1 (PRKAA1) is the catalytic subunit of AMP-activated protein kinase, acting as a key cellular energy sensor. Upon reduction of intracellular ATP levels, it activates energy-producing pathways and inhibits energy-consuming processes such as protein, carbohydrate, and lipid biosynthesis. Related references: PMID:17307971 PMID:17712357
- Lipid and cholesterol metabolism regulation: It directly phosphorylates and inactivates acetyl-CoA carboxylase (ACACA/ACACB) and HMG-CoA reductase (HMGCR), thereby suppressing fatty acid and cholesterol synthesis. Related references: PMID:17307971 PMID:17712357
- Transcription regulation: AMPKα1 modulates expression of genes involved in energy metabolism by phosphorylating transcription regulators such as CRTC2, FOXO3, HDAC5, MEF2C, and SREBF1. Related references: PMID:11518699 PMID:11554766 PMID:15866171
- mTORC1 and autophagy control: In response to nutrient limitation, AMPKα1 phosphorylates TSC2 and RPTOR to negatively regulate the mTORC1 complex, and phosphorylates ULK1 to initiate autophagy. Related references: PMID:14651849 PMID:18439900 PMID:20160076 PMID:21205641 PMID:28561066
- Subcellular distribution: AMPKα1 is found in the cytoplasm, nucleus, and late endosomes, consistent with its pleiotropic regulatory roles.
- Kinase properties: As a serine/threonine-protein kinase, it binds ATP and Mg²⁺ for catalytic activity and is subject to phosphorylation and ubiquitination, reflecting tight post-translational control.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 251–550 of human AMPKα1 (Q13131); consider this region when designing epitope-specific detection strategies.
- For Western blot (WB), bands are expected at approximately 64 kDa (AMPKα1) and 65 kDa (AMPKα2); validate with appropriate molecular weight markers and positive/negative cell lysates.
- In immunohistochemistry (IHC-P), both cytoplasmic and nuclear staining may be observed; optimize antigen retrieval conditions and antibody titration on tissue microarrays.
- For ELISA applications, verify optimal coating concentration and buffer conditions using recombinant AMPK protein standards.
