Biological Background: AMOTL2 Function and Localization
- AMOTL2 (Angiomotin-like protein 2), also known as Leman coiled-coil protein (LCCP), is a multi-domain adaptor protein belonging to the angiomotin family.
- AMOTL2 regulates actin cytoskeleton architecture, anchoring radial actin fibers to CDH1-based adherens junctions and enabling cellular response to contractile forces, thereby contributing to epithelial sheet integrity (PubMed:28842668). Related references: PMID:28842668
- It inhibits Wnt/β-catenin signaling by promoting CTNNB1 sequestration in recycling endosomes, and activates the Hippo pathway via WWP1-mediated ubiquitination, which recruits LATS2 to phosphorylate YAP1, leading to its cytoplasmic retention and inhibition of YAP1-driven transcription (PubMed:34404733, PubMed:17293535, PubMed:21205866, PubMed:26598551). - The immunogen corresponds to the C-terminal region (aa 638–837) of human AMOTL2; the antibody may detect full-length and potential isoforms or degradation products.
- For WB, expect a band at ~86 kDa; consider using endothelial (e.g., HUVEC) or epithelial cell lysates as positive controls. Both human and mouse samples are reported cross-reactive.
