Biological Background: AMHR2 Function and Localization
- AMHR2 encodes the anti-Muellerian hormone type-2 receptor (also known as AMH type II receptor, MIS type II receptor; aliases MISRII, MRII), a transmembrane serine/threonine kinase.
- The protein is a key component of the AMH signaling pathway, mediating Müllerian duct regression in male sexual differentiation.
- Upon AMH binding, two type II receptors and two type I receptors form a heterotetrameric complex, where type II receptors phosphorylate and activate type I receptors, which then phosphorylate SMAD transcriptional regulators to modulate gene expression.
- AMHR2 is localized to the plasma membrane (membrane-bound receptor).
- The receptor contains intrinsic kinase activity, requiring ATP and divalent cations (Mg²⁺/Mn²⁺) for function, and undergoes post-translational modifications including glycosylation and disulfide bond formation.
- Alternative splicing generates multiple isoforms; mutations in AMHR2 are linked to persistent Müllerian duct syndrome (PMDS), a form of pseudohermaphroditism.

- This antibody is validated for WB and ELISA; for WB, a band around 52-63 kDa (likely corresponding to glycosylated or alternatively spliced isoforms) may be observed.
- For ELISA, consider coating with recombinant AMHR2 protein or peptide to establish standard curves.
- Since cross-reactivity is confirmed only for human, validate species specificity if used with other samples.
