Biological Background: AMBP Function and Localization

  • The AMBP gene encodes a precursor protein that is cleaved into two functional chains: alpha-1-microglobulin (protein HC/A1M) with antioxidant and tissue repair activities, and bikunin, a kunitz-type serine protease inhibitor.
  • Alpha-1-microglobulin (A1M) exhibits reductase, heme-binding, and radical-scavenging activities. It reduces extracellular methemoglobin, binds and degrades free heme to prevent hemolysis, and protects against oxidative damage to low-density lipoprotein particles and extracellular matrix structures. Subcellularly, it localizes to the extracellular space, endoplasmic reticulum, cytoplasm, cell membrane, nucleus membrane, and mitochondrial inner membrane.

Experimental Guidance and Technical Tips

  • The immunogen used spans amino acids 20-202 of human AMBP, covering the alpha-1-microglobulin (A1M) chain. The antibody is expected to recognize A1M and may also detect the full-length precursor; consider this when interpreting bands in Western blot.
  • In WB, the full-length AMBP has a predicted molecular weight of ~39 kDa; however, processed chains or glycosylated forms may appear at different sizes. Use appropriate molecular weight markers and consider reducing vs. non-reducing conditions.
  • For ELISA applications, this polyclonal antibody can be used as a detection reagent. If a sandwich ELISA format is desired, pairing with a monoclonal capture antibody specific to a different epitope may improve assay specificity.
  • The antibody cross-reacts with mouse AMBP, indicating potential epitope conservation. Validation in mouse-derived samples is recommended before use in mouse experimental systems.
  • Due to the presence of disulfide bonds in bikunin, non-reducing conditions may be necessary to preserve epitope recognition if targeting the bikunin chain.
