Biological Background: AMACR Function and Localization

  • Alpha-methylacyl-CoA racemase (AMACR), also known as 2-methylacyl-CoA racemase, is encoded by the AMACR gene (Gene ID: 23600) and is essential for branched-chain fatty acid metabolism.
  • AMACR catalyzes the racemization of alpha-methyl-branched-chain fatty acyl-CoA esters, converting (R)- and (S)-stereoisomers, which is critical for the beta-oxidation of branched-chain fatty acids in peroxisomes. Related references: PMID:10655068 PMID:11060359 PMID:7649182
  • The enzyme acts exclusively on coenzyme A thioesters and accepts substrates including pristanoyl-CoA and trihydroxycoprostanoyl-CoA (involved in bile acid synthesis), as well as the arylpropionic acid ibuprofen, but does not act on 3-methyl-branched or linear-chain acyl-CoAs. Related references: PMID:10655068 PMID:11060359 PMID:7649182
  • AMACR localizes to both peroxisomes and mitochondria, consistent with its role in organellar fatty acid metabolism.
  • Post-translational modifications include acetylation, and the gene is subject to alternative splicing (UniProt keyword: Acetylation, Alternative splicing).
  • Genetic variants in AMACR are linked to intrahepatic cholestasis and other metabolic disorders (UniProt keyword: Disease variant, Intrahepatic cholestasis).

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 1-138 of human AMACR) targets the N-terminal region; consider potential epitope masking or absence in some splice variants and validate in the relevant sample system.
  • For Western blot, a band near 42 kDa is expected; optimize antibody dilution and exposure settings, and confirm reactivity with mouse or rat tissue lysates if cross-species studies are planned.
  • In ELISA applications, the recombinant immunogen can serve as a positive control; assess coating and detection conditions for the target antigen in biological matrices.
  • Due to peroxisomal and mitochondrial localization, subcellular fractionation or organelle enrichment may improve detection sensitivity in complex samples.
