Biological Background: Alpha-methylacyl-CoA racemase Function and Localization
- Alpha-methylacyl-CoA racemase (AMACR), also termed 2-methylacyl-CoA racemase, is an enzyme with dual localization in peroxisomes and mitochondria.
- It catalyzes the interconversion of (R)- and (S)-stereoisomers of alpha-methyl-branched-chain fatty acyl-CoA esters, essential for the degradation of branched-chain fatty acids. Substrates include pristanoyl-CoA, trihydroxycoprostanoyl-CoA (intermediate in bile acid synthesis), and the drug ibuprofen. Related references: PMID:7649182, PMID:10655068, PMID:11060359
- AMACR acts exclusively on coenzyme A thioesters and does not process free fatty acids or 3-methyl-branched/linear-chain acyl-CoAs.
- The enzyme is encoded by the AMACR gene, which undergoes alternative splicing to produce multiple protein isoforms.
- Post-translational modifications include acetylation, which may regulate its activity or stability.
- Defects in AMACR are associated with intrahepatic cholestasis and other metabolic conditions, highlighting its clinical significance.
Experimental Guidance and Technical Tips
- The immunogen used was a recombinant protein corresponding to amino acids 1–138 of human AMACR (NP_055139.4). This N-terminal region is likely to generate antibodies recognizing multiple isoforms.
- For Western blot (WB), a band around 42 kDa is expected based on the predicted molecular weight.
- Immunofluorescence (IF) and immunohistochemistry (IHC) can be employed to visualize AMACR localization in cells and tissues; consider including appropriate organelle markers to confirm peroxisomal and mitochondrial distribution.
- Given the high sequence conservation among human, mouse, and rat AMACR, this antibody is predicted to detect the protein in these species; however, validation in the relevant species and sample type is recommended.
- The antibody may be suitable for ELISA applications; however, appropriate assay optimization and controls are necessary.
