Biological Background: Alpha-synuclein Function and Localization

  • Alpha-synuclein is a neuronal protein encoded by the SNCA gene (also known as NACP or PARK1) and is a member of the synuclein family.
  • It plays critical roles in synaptic activity, including the regulation of synaptic vesicle trafficking and neurotransmitter release. Related references: PMID:20798282 PMID:26442590 PMID:28288128
  • Functions as a monomer to enhance synaptic vesicle exocytosis by increasing local Ca2+ release from microdomains. Related references: PMID:30404828
  • Acts as a molecular chaperone in its multimeric membrane-bound state, assisting in the folding of SNARE proteins to sustain SNARE-complex assembly during aging. Related references: PMID:20798282
  • Associates with the dopamine transporter (DAT1) to modulate dopamine neurotransmission. Related references: PMID:26442590
  • Subcellularly localized to the cytoplasm, membrane, nucleus, synapse, and is secreted; also found in cell projections including axons.
  • Highly expressed in presynaptic terminals of the central nervous system, principally in brain.
  • Post-translationally modified by acetylation, phosphorylation, and ubiquitylation; implicated in neurodegenerative diseases such as Parkinson's and Alzheimer's.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 59–140 of human alpha-synuclein, which includes the hydrophobic NAC domain; this region is conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
  • For Western blotting, consider using brain tissue lysates to detect the endogenous protein at ~14 kDa.
  • For immunofluorescence or immunohistochemistry, take into account alpha-synuclein's multiple subcellular localizations (cytoplasm, membrane, nucleus, synapse, axon) and validate staining in relevant neuronal cell lines or brain sections.
  • ELISA applications may benefit from using recombinant alpha-synuclein as a standard; ensure appropriate buffer conditions for monomeric or oligomeric forms as needed.
