Biological Background: Alpha Internexin Function and Localization
- Alpha-internexin (INA), also known as neurofilament 5 (NEF5), NF-66, or 66 kDa neurofilament protein, is a class-IV neuronal intermediate filament protein.
- It self-assembles and is involved in neuronal morphogenesis; it may form independent structural networks or cooperate with NEFL to create a filamentous backbone to which NEFM and NEFH attach.
- May also interact with the neuronal intermediate filament protein peripherin (PRPH) to form filamentous networks.
- Expression is predominantly found in the adult central nervous system (CNS).
- It is annotated with key post-translational modifications including acetylation and phosphorylation, and is classified as a glycoprotein.
- The protein contains a coiled-coil domain and plays roles in neurogenesis and neuronal differentiation.
- Alpha-internexin has been identified through direct protein sequencing and proteomics approaches.
Experimental Guidance and Technical Tips
- This antibody is raised against a synthetic peptide spanning amino acids 350-450 of human Alpha Internexin; peptide blocking experiments can be used to confirm specificity in your assay system.
- For Western blot, the predicted molecular weight is approximately 55 kDa; consider running appropriate positive controls from CNS tissue lysates.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), validation in human, mouse, or rat CNS tissues is recommended given the predominant expression in adult brain.
- Cross-reactivity with mouse and rat suggests utility in rodent model studies, but optimal working dilutions should be empirically determined.
