  • Alpha-fetoprotein (AFP), also referred to as alpha-1-fetoprotein or alpha-fetoglobulin, is encoded by the human AFP gene.
  • AFP belongs to the albumin gene family and shares structural features with serum albumin.
  • This secreted protein is synthesized by the fetal liver and yolk sac, and circulates in fetal plasma.
  • AFP functions as a carrier protein, binding copper, nickel, fatty acids, and, to a lesser extent, bilirubin. A small proportion (<2%) interacts with estrogen.
  • Post-translational modifications include glycosylation, phosphorylation, disulfide bond formation, and sulfation; the protein's three-dimensional structure has been solved.
  • Due to its fetal origin and secretion into plasma, AFP levels are developmentally regulated.
