Biological Background: alpha + beta Synuclein Function and Localization
- Alpha-synuclein (SNCA) is a neuronal protein highly expressed in presynaptic terminals in the central nervous system, principally in the brain.
- It regulates synaptic vesicle trafficking and neurotransmitter release by enhancing vesicle priming, fusion, and dilation of exocytotic fusion pores, partly via increasing local Ca²⁺ release.
Related references: PMID:28288128, PMID:30404828 - In its multimeric membrane-bound state, alpha-synuclein acts as a molecular chaperone, assisting in the folding of SNARE proteins at the presynaptic plasma membrane in conjunction with cysteine string protein-alpha/DNAJC5, and sustaining SNARE-complex assembly during aging.
Related references: PMID:20798282 - It modulates dopamine neurotransmission by associating with the dopamine transporter (DAT1), thereby affecting its activity.
Related references: PMID:26442590 - Subcellularly, alpha-synuclein localizes to the cytoplasm, membrane, nucleus, synapse, and is secreted; it is also found in cell projections including axons.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitination; the protein binds metal ions such as copper.
- Alpha-synuclein is a major component of amyloid plaques in Alzheimer’s disease, and genetic variants are linked to Parkinson disease and parkinsonism.
- The antibody also recognizes beta-synuclein (SNCB), a homologous protein that shares sequence similarity with alpha-synuclein and is often co-expressed in neuronal tissues.
Experimental Guidance and Technical Tips
- For Western blot, consider that the observed molecular weight (~18 kDa) may be higher than the predicted 14.4 kDa due to post-translational modifications; validate using appropriate positive controls (e.g., brain lysates).
- In immunofluorescence (IF-P), permeabilization is typically required; optimize fixation and permeabilization conditions for the specific cell type or tissue.
- For ELISA, the antibody may be used as capture or detection reagent; titration and pairing with a suitable detection antibody are recommended.
- Cross-reactivity with mouse and rat synucleins has been confirmed; however, verify performance in your experimental system with appropriate species controls.
