Biological Background: Alpha-2-Macroglobulin (A2M) Function and Localization
- Alpha-2-macroglobulin (A2M) is a large plasma glycoprotein and member of the alpha-macroglobulin family, also known as C3 and PZP-like alpha-2-macroglobulin domain-containing protein 5 (CPAMD5).
- A2M acts as a broad-spectrum protease inhibitor, capable of inhibiting all four classes of proteinases (serine, cysteine, aspartic, and metalloproteinases) through a unique 'trapping' mechanism.
- The protein contains a 'bait region' with specific cleavage sites; upon proteolytic cleavage, a conformational change traps the proteinase, and a thioester bond mediates covalent binding to the enzyme.
- Following cleavage, the entrapped proteinase remains active against low molecular weight substrates but shows greatly reduced activity against high molecular weight substrates.
- A2M is secreted into the blood plasma and functions as a key extracellular protease scavenger, contributing to the regulation of coagulation, fibrinolysis, and inflammation.
