  • Alpha-2-macroglobulin (A2M) is a large plasma glycoprotein and member of the alpha-macroglobulin family, also known as C3 and PZP-like alpha-2-macroglobulin domain-containing protein 5 (CPAMD5).
  • A2M acts as a broad-spectrum protease inhibitor, capable of inhibiting all four classes of proteinases (serine, cysteine, aspartic, and metalloproteinases) through a unique 'trapping' mechanism.
  • The protein contains a 'bait region' with specific cleavage sites; upon proteolytic cleavage, a conformational change traps the proteinase, and a thioester bond mediates covalent binding to the enzyme.
