Biological Background: ALOXE3 Function and Localization
- ALOXE3 is a non-heme iron-containing lipoxygenase that functions primarily as a hydroperoxide isomerase, converting fatty acid hydroperoxides into epoxyalcohols and ketones. Related references: PMID:12881489, PMID:17045234, PMID:20923767
- In the skin, it processes linoleate in esterified omega-hydroxyacyl-sphingosine ceramides, generating an epoxy-ketone derivative essential for covalent attachment to membrane proteins, thus forming the corneocyte lipid envelope and establishing the skin water barrier. Related references: PMID:21558561
- ALOXE3 also exhibits residual lipoxygenase activity, oxygenating polyunsaturated fatty acids like arachidonic acid to produce hydroperoxides. Related references: PMID:20921226
- Through hepoxilin production, it may contribute to adipocyte differentiation by activating PPARG and regulate inflammatory pain signaling in the spinal cord.
- The protein is predominantly expressed in the skin and localizes to the cytoplasm.
- Keywords including alternative splicing suggest multiple isoforms, and mutations in ALOXE3 are linked to ichthyosis, highlighting its role in epidermal homeostasis.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 1–280 amino acids; consider its coverage of known isoforms when interpreting results.
- For Western blotting, validate antibody performance in skin tissue lysates from human, mouse, or rat, given the skin-specific expression profile.
- For ELISA, optimize coating and detection conditions using purified recombinant protein or tissue extracts.
- As a cytoplasmic protein, prepare whole-cell or cytoplasmic fractions; avoid nuclear-enriched preparations.
- Polyclonal antibodies may exhibit lot-to-lot variability; include appropriate positive and negative controls in each experiment.
- Cross-reactivity with mouse and rat makes this antibody suitable for translational studies in rodent models.
