Biological Background: ALOX5 Function and Localization
- ALOX5 encodes polyunsaturated fatty acid 5-lipoxygenase (also known as arachidonate 5-lipoxygenase, 5-LO, 5-lipoxygenase), a key enzyme in the biosynthesis of leukotrienes, potent inflammatory mediators, via oxygenation of arachidonic acid. - It converts arachidonate to 5-hydroperoxyeicosatetraenoate (5-HPETE) and then to leukotriene A4 (LTA4), and can also produce 8-HPETE and 12-HPETE. Related references: PMID:23246375
- ALOX5 displays lipoxin synthase activity, converting (15S)-HETE into a conjugate tetraene, and metabolizes other polyunsaturated fatty acids to form specialized pro-resolving mediators (SPMs) like lipoxins and resolvins, contributing to anti-inflammatory responses. - It is a calcium- and iron-binding enzyme, classified as a dioxygenase and hydrolase, with alternative splicing and phosphorylation modifications.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–200) of human ALOX5; the antibody may detect full-length and possibly N-terminal fragments. Validate specificity in your sample system.
- Predicted band size is ~78 kDa in Western blot; consider using appropriate molecular weight markers and loading controls. Optimize lysis buffers and protease inhibitors for membrane and nuclear fractions.
- For immunohistochemistry (IHC‑P), test antigen retrieval methods (citrate or EDTA buffer) and antibody titration. Include relevant positive and negative tissue controls.
- For immunofluorescence (IF/ICC), permeabilization with Triton X‑100 may be necessary to access nuclear and cytoplasmic pools.
