Biological Background: ALOX15 Function and Localization

  • ALOX15 (12/15-lipoxygenase) is a non-heme iron-containing dioxygenase that catalyzes the stereo-specific peroxidation of free and esterified polyunsaturated fatty acids, generating bioactive lipid mediators such as 12-HPETE and 15-HPETE from arachidonate. Related references: PMID:17052953 PMID:1944593 PMID:24282679
  • It also peroxidizes linoleate to 13-hydroperoxyoctadecadienoate (13-HPODE) and converts epoxy fatty acids to monocyclic endoperoxides via hydroperoxy-epoxide intermediates. Related references: PMID:8334154 PMID:25293588
  • Participates in the synthesis of specialized pro-resolving mediators (SPMs) such as resolvin D5 from DHA, which actively down-regulate immune responses and inhibit platelet aggregation. Related references: PMID:32404334
  • Plays a role in immune self-tolerance by peroxidizing membrane phosphatidylethanolamine to signal the clearance of apoptotic cells, and contributes to epithelial wound healing in the cornea through lipoxin A4 and neuroprotectin D1 production.
  • Involved in the cellular response to interleukin-13 (IL-13), linking lipoxygenase activity to cytokine signaling. Related references: PMID:21831839
  • Subcellular localization includes cytosol, cell membrane, and lipid droplets, reflecting its dynamic role in lipid metabolism and signaling.
  • Detected in monocytes, eosinophils, and airway epithelial cells, consistent with its involvement in innate immunity and airway inflammation.
  • Multiple isoforms exist due to alternative splicing; catalytic activity requires non-heme iron and calcium binding.

Experimental Guidance and Technical Tips

  • For Western blot, use reducing conditions and expect a band near 75 kDa. Validate expression in relevant human cell lines (e.g., THP-1 monocytes, A549 airway epithelial cells) or tissues expressing ALOX15.
  • For immunohistochemistry (paraffin), consider antigen retrieval and use appropriate positive controls (e.g., inflamed lung or atherosclerotic lesion sections containing macrophages).
  • In ELISA applications, pair with a matched detection antibody; use recombinant ALOX15 protein as a standard and include proper controls for cross-reactivity with mouse and rat samples.
  • Given cross-reactivity with mouse and rat, verify specificity in each species before quantitative assays; perform sequence alignment if necessary.
