Biological Background: ALKBH8 Function and Localization

  • ALKBH8 is a multifunctional enzyme that acts as a tRNA (carboxymethyluridine(34)-5-O)-methyltransferase and an alpha-ketoglutarate-dependent dioxygenase, also known as ABH8, TRM9, or TRMT9A.
  • It catalyzes the methylation of 5-carboxymethyl uridine to 5-methylcarboxymethyl uridine at the wobble position of tRNA anticodon loops, preferentially targeting tRNA(Arg) and tRNA(Glu). Related references: PMID:20123966 PMID:20308323 PMID:31079898
  • Through its dioxygenase domain, ALKBH8 hydroxylates 5-methylcarboxymethyl uridine to 5-(S)-methoxycarbonylhydroxymethyluridine, with a preference for tRNA(Gly). Related references: PMID:21285950
  • The protein is required for normal survival after DNA damage and may inhibit apoptosis while promoting cell survival and angiogenesis. Related references: PMID:20308323 PMID:19293182
  • ALKBH8 is localized in both the cytoplasm and nucleus, and is widely expressed with highest levels in spleen, pancreas, and lung.

  • For Western blot, a band around 75 kDa is expected based on the predicted molecular weight; validate in your sample lysate and optimize blocking/detection conditions.
  • This antibody has been tested in IHC-P and ELISA; for new sample types or applications, perform pilot experiments and include appropriate controls.
  • Cross-reactivity with human, mouse, and rat has been reported, but homology with other species should be checked if used in non-listed organisms.
