Biological Background: ALKBH5 Function and Localization

  • Functions as an RNA N6-methyladenosine (m6A) demethylase, removing m6A marks from mRNA through an oxidative mechanism that requires molecular oxygen, alpha-ketoglutarate, and iron. Related references: PMID:21264265 PMID:23177736 PMID:24489119
  • m6A demethylation by ALKBH5 affects mRNA processing, translation, and nuclear export, thereby regulating gene expression. Related references: PMID:23177736 PMID:34048572 PMID:36944332
  • In vitro, ALKBH5 can also demethylate N6-methyladenosine in single-stranded DNA. Related references: PMID:24616105
