Biological Background: ALKBH3 Function and Localization
- ALKBH3, also known as prostate cancer antigen-1 (PCA1) or hABH3, is an alpha-ketoglutarate-dependent dioxygenase belonging to the AlkB family of DNA/RNA repair enzymes, with alternative splicing and ubiquitin conjugation modifications reported.
- It catalyzes oxidative demethylation of 1-methyladenosine (m1A) and 3-methylcytosine (m3C) in both DNA and RNA, requiring molecular oxygen, alpha-ketoglutarate, and iron as cofactors.
Related references: PMID:12486230, PMID:12594517, PMID:16174769, PMID:16858410, PMID:22055184 - ALKBH3 preferentially acts on single-stranded DNA, but can process double-stranded substrates when recruited by the ASCC3 helicase, which unwinds DNA.
Related references: PMID:22055184 - The enzyme also demethylates N1-methyladenosine (m1A) in mRNA, particularly enriched in 5'-untranslated regions (UTRs) and near start codons, thereby regulating mRNA translation and stability.
Related references: PMID:26863196, PMID:26863410 - ALKBH3 localizes to both the nucleus and the cytoplasm, consistent with its dual functions in nuclear DNA repair and cytoplasmic RNA processing.
- It is ubiquitously expressed, with detection across heart, pancreas, skeletal muscle, thymus, testis, ovary, prostate, colon, and other tissues.
Experimental Guidance and Technical Tips
- The immunogen covers the near full-length protein (aa 1–286), suggesting that the polyclonal antibody likely recognizes multiple epitopes and can detect both native and denatured ALKBH3 in WB under reducing conditions, with an expected band around 33 kDa.
- For Western blot, consider using whole-cell lysates from human or mouse cell lines expressing ALKBH3; validate in your specific sample system.
- This antibody is also suitable for ELISA; optimize coating antigen and antibody concentration based on your experimental setup.
