Biological Background: ALKBH1 Function and Localization
- ALKBH1 is a nucleic acid dioxygenase that acts on DNA and tRNA, requiring molecular oxygen, alpha-ketoglutarate, and iron as cofactors. Related references: PMID:18603530 PMID:27497299
- It primarily functions as a tRNA demethylase, removing N1-methyladenine from various tRNAs, with preference for position 58 (m1A58) in the T-loop, thereby regulating translation initiation and elongation. Related references: PMID:27745969
- In mitochondria, ALKBH1 oxidizes mt-tRNA(Met) at cytosine(34) to 5-formylcytosine, expanding codon recognition to read AUA in addition to AUG. Related references: PMID:27497299
