Biological Background: ALKBH1 Function and Localization

  • ALKBH1 is a nucleic acid dioxygenase that acts on DNA and tRNA, requiring molecular oxygen, alpha-ketoglutarate and iron. Related references: PMID:18603530, PMID:27497299
  • Primarily functions as a tRNA demethylase, removing N1-methyladenine (m1A) from tRNAs, especially at position 58 (m1A58), regulating translation initiation and elongation in response to glucose deprivation. Related references: PMID:27745969
  • In mitochondria, ALKBH1 oxidizes mt-tRNA(Met) at cytosine 34 to form 5-formylcytosine (f5C), expanding codon recognition for AUA/AUG. Related references: PMID:27497299
