Biological Background: ALKBH1 Function and Localization
- ALKBH1 is a nucleic acid dioxygenase that acts on DNA and tRNA, requiring molecular oxygen, alpha-ketoglutarate and iron. Related references: PMID:18603530, PMID:27497299
- Primarily functions as a tRNA demethylase, removing N1-methyladenine (m1A) from tRNAs, especially at position 58 (m1A58), regulating translation initiation and elongation in response to glucose deprivation. Related references: PMID:27745969
- In mitochondria, ALKBH1 oxidizes mt-tRNA(Met) at cytosine 34 to form 5-formylcytosine (f5C), expanding codon recognition for AUA/AUG. Related references: PMID:27497299

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–100), suggesting recognition of the full-length native protein and potentially certain isoforms containing this region. Validate specificity in the relevant sample system.
- For Western blotting, expect a band at approximately 44 kDa. Use appropriate loading controls and confirm cross-reactivity with human, mouse, and rat lysates.
- In immunoprecipitation (IP), pre-clearing and optimized antibody-to-lysate ratios are recommended to minimize non-specific binding.
- For immunohistochemistry (IHC-P), antigen retrieval conditions may need optimization due to the nuclear/mitochondrial localization. Consider testing both standard citrate and EDTA-based retrieval.
