Biological Background: ALKBH1 Function and Localization
- ALKBH1 is a nucleic acid dioxygenase dependent on Fe(II), α-ketoglutarate, and molecular oxygen, and is the human homolog of E. coli AlkB.
Related references: PMID:18603530, PMID:27497299 - It mediates the demethylation of N(1)-methyladenine (m1A) at position 58 of various tRNAs, regulating translation initiation and elongation in response to glucose deprivation.
Related references: PMID:27745969 - In mitochondria, ALKBH1 specifically oxidizes 5-methylcytosine (m5C) at position 34 of mt-tRNA(Met) to 5-formylcytosine (f5C), expanding codon recognition during mitochondrial translation.
Related references: PMID:27497299 - ALKBH1 serves as a DNA N6-methyladenine (6mA) demethylase at L1 retrotransposons in embryonic stem cells, potentially contributing to epigenetic silencing.
Related references: PMID:30017583, PMID:30392959 - It also demethylates N(3)-methylcytidine (m3C) in mRNA, broadening its role in RNA modification.
Related references: PMID:31188562 - Additionally, ALKBH1 has DNA lyase activity that cleaves DNA at abasic sites independently of its dioxygenase function, generating double-strand breaks.
Related references: PMID:19959401, PMID:23577621 - ALKBH1 localizes to both the nucleus and mitochondria, compartmentalizing its diverse nucleic acid modification functions.
- Ubiquitously expressed across human tissues, ALKBH1 may also play a role in placental trophoblast lineage differentiation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 290-389 of human ALKBH1. Alternative immunogens targeting other regions may yield antibodies with different epitope specificities.
- For Western blotting, a band corresponding to the predicted molecular weight of ~44 kDa is expected under reducing conditions. Verify band specificity using appropriate controls.
- In immunoprecipitation, the antibody can potentially capture human, mouse, and rat ALKBH1 proteins; optimize the antibody-to-lysate ratio.
- For immunofluorescence/ICC, co-localization studies with organelle-specific markers can confirm the nuclear and mitochondrial distributions of ALKBH1.
- ELISA applications may benefit from using recombinant ALKBH1 protein or peptide as the capture antigen; titrate the antibody for optimal sensitivity.
