Biological Background: Alkaline Phosphatase (ALPL) Function and Localization

  • Alkaline phosphatase, tissue-nonspecific isozyme (ALPL), also known as liver/bone/kidney isozyme, exists in multiple isoforms due to alternative splicing and is widely expressed in bone, liver, and kidney tissues.
  • Plays an essential role in skeletal and dental mineralization by hydrolyzing extracellular diphosphate (PPi), a potent mineralization inhibitor, into phosphate, thereby promoting hydroxyapatite crystal formation and increasing inorganic phosphate concentration.   • The immunogen corresponds to amino acids 230-440 of human ALPL, within the mature protein's central region. Because ALPL undergoes alternative splicing, ensure the epitope is present in your isoform of interest.
  • For Western blot (WB), the predicted molecular weight is approximately 57 kDa; however, glycosylation and other post-translational modifications may cause the observed band to shift to a higher apparent molecular weight. Use positive control lysates from bone, liver, or kidney tissues, or ALPL-overexpressing cell lines.
  • For immunohistochemistry (IHC-P), optimize antigen retrieval conditions (e.g., heat-induced epitope retrieval) and validate staining specificity using ALPL-deficient or knockout tissue controls if available.
  • For ELISA, the antibody can be used as capture or detection reagent; titrate antibody concentrations against recombinant ALPL protein or natural samples to determine optimal working dilutions.
